目的:从血栓素(TXA_2)与前列环素(PGI_2)平衡、肿瘤坏死因子(TNF)、多巴胺(DA)、去甲肾上腺素(NE)及肾上腺素(E)含量的变化探讨益元活血丹和大黄及血栓心脉宁对老龄大鼠脑缺血再灌注肺脏损伤保护作用的机制。方法:老龄(20月龄以上)大鼠分为老龄对照组,老龄模型组,尼莫地平组,益元活血丹大、小剂量组,大黄组及血栓心脉宁组,观察大鼠全脑缺血30 min 再灌注60 min 后肺脏组织中 TXB_2、6-Keto-PGF_(1α)、TNF 和血清及下丘脑 DA、NE、E 含量的变化。结果:老龄模型组肺组织 TXB_2/6-Keto-PGF_(1α)比值降低,TNF 含量增加,与老龄对照组比较差异有显著性(P 均<0.05),老龄模型组大鼠交感-肾上腺系统兴奋增强,与老龄模型组比较,益元活血丹大剂量组 TXB_2/6-Keto-PGF_(1α)比值提高(P<0.01),血栓心脉宁组、大黄组、尼莫地平组肺组织 TNF 含量降低(P<0.05或 P<0.01);用药各组交感-肾上腺系统兴奋减低,以大黄组和益元活血丹大、小剂量组明显。结论:TXA_2与 PGI_2平衡失调、TNF 增高及交感-肾上腺系统兴奋增强可能参与老龄大鼠脑缺血再灌注肺损伤。大黄和益元活血丹及血栓心脉宁对肺损伤保护作用机制可能与其抑制交感-肾上腺系统兴奋、拮抗 TNF 损伤及调节 TXA_2与 PGI_2平衡失调有关,各组药物的作用有所侧重。
Objective: To explore the changes of the contents of TXA_2 and prostacyclin (PGI_2), tumor necrosis factor (TNF), dopamine (DA), norepinephrine (NE) and epinephrine (E) in Yiyuan Huoxuedan Mechanism of Protective Effect of Dahuang Youshuan Xinmaining on Lung Injury of Cerebral Ischemia Reperfusion in Aged Rats . METHODS: Older age (20 months or older) rats were divided into old age control group, old age model group, nimodipine group, Yiyuan Huoxue Danda, low dose group, rhubarb group and Xueshuan Xinmaining group. Changes in TXB_2, 6-Keto-PGF_(1α), TNF, and serum levels of DA, NE, and E in the hypothalamus after 30 minutes of ischemia and 60 minutes after reperfusion. Results: The ratio of TXB_2/6-Keto-PGF_(1α) in lung tissue of aged model group was decreased and the TNF content was increased. There was significant difference compared with the aged control group (P<0.05). The sympathetic-adrenal system was excited in the aged model rats. Compared with the aged model group, the ratio of TXB_2/6-Keto-PGF_(1α) in the high-dose group of Yiyuan Huoxuedan increased (P<0.01), and the TNF content in the lungs of Xuetong Xinmaining group, Dahuang group and Nimodipine group was increased. Decreased (P<0.05 or P<0.01); The sympathetic-adrenal system excitement was reduced in each group, and it was evident in the rhubarb group and the Yiyuan Huoxue Dan large and low dose groups. Conclusion: The imbalance of TXA_2 and PGI_2, TNF elevation and sympathetic-adrenal system stimulatory enhancement may be involved in cerebral ischemia-reperfusion lung injury in aged rats. The protective mechanism of Dahuang Huoxue Huoxue Dan and Xueshuan Xinmaining on lung injury may be related to its inhibition of sympathetic-adrenal system excitement, antagonism of TNF injury, and regulation of imbalance between TXA 2 and PGI 2 balance. The effects of each group of drugs have been emphasized.
