目的:探讨磁共振导引下注射rAd-p53治疗兔脑VX2瘤的可行性研究。方法:12只新西兰大白兔随机分为对照组(A组)、治疗组(B组)两组,A、B组各为6只,全麻下通过颅骨钻孔种植瘤块的方法建立VX2移植性脑瘤模型。脑瘤生长至直径平均约0.5cm时,在磁共振导引下穿刺肿瘤并分别靶向注射0.5ml无菌生理盐水(A组)和0.5ml rAd-p53(B组),随后各组分别在术后1天、3天、5天行MRI扫描后每组处死2只,行病理学及免疫荧光检查。结果:12只兔全部有脑瘤生长,且肿瘤大小较一致,形态规则,MR扫描呈T1WI略低信号,T2WI呈略高信号;增强扫描(钆喷替酸葡甲胺0.1mmol/kg体重)呈明显较均匀强化,肿瘤最大直径0.5~0.7cm,和病理学表现相一致,能够满足介入治疗的需要。所有注射无菌生理盐水和rAd-p53的介入操作均成功进行,术后1、3、5天病理学及MRI均未观察到明显差异,但在免疫荧光上,B组P53蛋白表达及凋亡均明显强于A组。结论:MRI导引下脑瘤注射rAd-p53治疗方法可行,能提高P53蛋白表达率及肿瘤凋亡率。
Objective: To investigate the feasibility of magnetic resonance guided injection of rAd-p53 in rabbit brain VX2 tumors. Methods: Twelve New Zealand white rabbits were randomly divided into control group (group A), treatment group (group B), and group A and group B (6 rabbits each). When the brain tumor grew to an average diameter of about 0.5 cm, the tumor was punctured by magnetic resonance and injected with 0.5 ml of sterile physiological saline (group A) and 0.5 ml of rAd-p53 (group B) respectively. Then, After 1 day, 3 days and 5 days after operation, 2 rats in each group were sacrificed and their pathology and immunofluorescence were performed. Results: All 12 rabbits had brain tumor growth, and the tumors were more consistent in size and morphology. MR scan showed a slightly lower signal of T1WI and a slightly higher signal of T2WI. Enhanced scanning (gadolinium doxorubicin 0.1 mmol / kg body weight) Was significantly more uniform enhancement, the largest tumor diameter 0.5 ~ 0.7cm, consistent with the pathological findings, to meet the needs of interventional therapy. All the injection of sterile saline and rAd-p53 intervention were successfully performed, no significant differences in pathology and MRI were observed on the 1st, 3rd and 5th postoperative days, but on immunofluorescence, P53 protein expression and apoptosis Were significantly stronger than the A group. Conclusion: MRI-guided brain tumor injection of rAd-p53 treatment is feasible, can increase the P53 protein expression rate and tumor apoptosis rate.
