目的 分析原发性非小细胞肺癌(NSCLC) 中p53 基因第5 ~8 外显子点突变发生及其意义。方法 运用聚合酶链反应- 单链构象多态性分析方法(PCRSSCP 分析法) 进行检测。结果 发现8 例肺良性疾病均无点突变发生,40 例NSCLC 中19例(47 .5 % ) 有点突变发生,点突变发生与性别,年龄,组织学类型和组织学分级无明显相关性( P> 0 .05) ,但与临床分期和淋巴结累及状态明显相关( P< 0 .01 ,P< 0 .05) 。结论 结果表明p53 基因点突变在NSCLC 的发生和进展中起着相当重要的作用。
Objective To analyze the occurrence and its significance of point mutations in exon 5 to exon 8 of p53 gene in primary non-small cell lung cancer (NSCLC). Methods Polymerase chain reaction-single strand conformation polymorphism (PCR-SSCP analysis) was used to detect. There was no point mutation in 8 cases of benign lung diseases. There were some mutations in 19 cases (47.5%) of 40 cases of NSCLC. There was no significant correlation between point mutation and gender, age, histological type and histological grade > 0 .05), but significantly correlated with clinical stage and lymph node involvement (P <0.01, P <0.05). Conclusion The results show that p53 gene point mutation plays a very important role in the occurrence and progression of NSCLC.
