目的:通过研究摘除卵巢致肾虚骨质疏松症大鼠血清中GPR48、ATF4、BSP的蛋白表达,探讨摘除卵巢致肾虚骨质疏松症的病理机制,并研究左归丸防治疗效及其作用机制。方法:摘除雌性大鼠双侧卵巢的方法复制肾虚骨质疏松症模型,采用具有补肾作用的左归丸对实验大鼠治疗12周,以盖天力片作为阳性对照组,正常大鼠、假手术组作为标准对照组,模型大鼠作为空白对照组,用ELISA法检测肾虚骨质疏松症大鼠血清GPR48、ATF4、BSP的蛋白表达。结果:ELISA方法检测表明:正常大鼠血清中存在GPR48、ATF4、BSP的蛋白表达。模空组大鼠血清中GPR48、ATF4、BSP的蛋白表达水平有明显的下降。左归丸组、盖天力组用药12周后,与模空组相比较,左归丸组可显著上调血清中GPR48、ATF4、BSP的蛋白表达水平。结论:1正常大鼠血清中存在GPR48、ATF4、BSP的蛋白表达;2血清中GPR48、ATF4、BSP的蛋白表达异常变化可能是导致骨质疏松症发生的机理之一;3左归丸能够调控GPR48、ATF4、BSP的蛋白表达,表明左归丸对骨质疏松症有一定的防治作用。
OBJECTIVE: To study the pathological mechanism of kidney deficiency osteoporosis in ovariectomized rats by studying the expression of GPR48, ATF4 and BSP in the serum of ovariectomized rats with osteoarthritis of the kidney and to study the curative effect and mechanism of Zuogui Pill. Methods: The ovariectomized female rats were duplicated the osteoporosis model of kidney deficiency. The Zuogui pill with kidney-invigorating effect was used to treat the experimental rats for 12 weeks. Giant-day tablets were taken as the positive control group. The normal rats The rats in the operation group served as the standard control group and the model rats served as the blank control group. The protein expressions of GPR48, ATF4 and BSP in serum of the rats with experimental renal osteoporosis were measured by ELISA. Results: The results of ELISA showed that the protein expression of GPR48, ATF4 and BSP existed in normal rat serum. The protein expression of GPR48, ATF4 and BSP in the model group was significantly decreased. Zuoguiwan group and GTI group could increase the protein expression of GPR48, ATF4 and BSP in Zuoguiwan group after 12 weeks. CONCLUSIONS: 1 There are GPR48, ATF4 and BSP protein expression in the serum of normal rats.2 The abnormal expression of GPR48, ATF4 and BSP in serum may be one of the mechanisms leading to osteoporosis.3 Zuogui pill can regulate GPR48, ATF4, BSP protein expression, indicating that Zuogui Wan on osteoporosis have a preventive effect.
