目的:观察泄浊益肾方对高尿酸血症肾损害大鼠肾脏的保护作用及作用机制。方法:利用腺嘌呤加乙胺丁醇复制高尿酸血症肾损害大鼠模型,实验分为正常对照组、模型对照组、中药高剂量组、中药中剂量组、中药低剂量组及西药(别嘌呤醇)对照组(n=10),检测血清尿酸、肌酐、尿素氮及观察肾组织病理变化。结果:中药各剂量组大鼠血尿酸明显下降,与模型对照组相比差异显著(P<0.01),尤中药中剂量组;中药各剂量组血肌酐、尿素氮均较模型对照组降低(P<0.01或P<0.05),尤中药高、中剂量组更为显著;中药各剂量组病理改变较轻,接近正常对照组。结论:泄浊益肾方能够降低高尿酸血症肾损害大鼠的血尿酸、肌酐、尿素氮,改善肾功能,对肾脏病理有修复保护作用,可防治高尿酸血症肾损害。
Objective: To observe the protective effect of Xiezhuo Yishen Recipe on kidney of rats with hyperuricemia and its mechanism. Methods: A rat model of hyperuricemia and renal damage was established by adenine and ethambutol. The experiment was divided into normal control group, model control group, high dose Chinese medicine group, medium dose Chinese medicine group, low dose Chinese medicine group and western medicine Allopurinol) control group (n = 10). Serum uric acid, creatinine and urea nitrogen were measured and pathological changes of renal tissues were observed. Results: Serum uric acid was significantly decreased in each dose group compared with the model control group (P <0.01), especially in the middle-dose group; serum creatinine and urea nitrogen in each dose group were lower than those in the model control group (P <0.01 or P <0.05), especially in the middle and high dose of traditional Chinese medicine. The pathological changes in each dose group of TCM were lighter and closer to the normal control group. Conclusion: Xiezhuyishen Decoction can reduce the blood uric acid, creatinine and urea nitrogen in rats with hyperuricemia and renal damage, improve renal function, repair and protect renal pathology, and prevent and treat renal damage caused by hyperuricemia.
