目的:研究支气管哮喘(哮喘)肺组织和哮喘病人外周血淋巴细胞端粒酶逆转录酶(TERT)mRNA和蛋白质的表达,并探讨其在哮喘发病机制中的作用及其临床意义。方法:用核酸原位杂交和免疫组织化学方法检测了6只豚鼠哮喘模型组织、6只正常对照组肺组织、16例哮喘发作期和 14例正常人外周血淋巴细胞 TERT mRNA和蛋白质的表达。结果:哮喘豚鼠肺组织中气道上皮细胞和淋巴小结,以及哮喘病人外周血淋巴细胞TERT mRNA和蛋白质表达强度显著高于正常对照组,哮喘病人外周血淋巴细胞 TERT mRNA和蛋白质表达的阳性指数与哮喘病人 FEV1/预计值、MVV/预计值比值呈显著负相关,与血清IgE水平呈显著正相关。结论:(1)TERT在哮喘豚鼠气道上皮、肺淋巴小结和人体外周血淋巴细胞中表达上调;(2)检测人体外周血液中淋巴细胞对TERT表达强度有望可以作为衡量哮喘病情的指标。
Objective: To study the expression of telomerase reverse transcriptase (TERT) mRNA and protein in peripheral blood lymphocytes of bronchial asthmatic (asthmatic) lung and asthmatic patients, and to explore its role in the pathogenesis of asthma and its clinical significance. Methods: The expression of TERT mRNA and protein in 6 guinea pig asthma model tissues, 6 normal control lung tissues, 16 asthma attack and 14 normal peripheral blood lymphocytes were detected by in situ hybridization and immunohistochemistry. Results: The TERT mRNA and protein expression of airway epithelial cells and lymphoid nodules in asthmatic guinea pigs and peripheral blood lymphocytes in asthmatic patients were significantly higher than those in normal controls. The positive index of TERT mRNA and protein expression in peripheral blood lymphocytes of asthmatic patients were significantly correlated with Asthma patients FEV1 / predicted value, MVV / predicted value was significantly negatively correlated, and serum IgE levels were significantly positively correlated. Conclusion: (1) TERT is upregulated in the airway epithelium, pulmonary nodules and human peripheral blood lymphocytes in asthmatic guinea pigs. (2) The detection of TERT expression in lymphocytes in human peripheral blood may be used as an indicator of asthma.
