目的 为研究乳腺肿瘤中胎盘型谷胱甘肽S -转移酶 (GST -π)mRNA表达与乳癌发生和耐药的相关性。方法 利用RT -PCR技术 ,分析 2 0例乳腺癌病人的 10 8个HE染色阴性淋巴结GST -πmRNA的表达。结果 GST-πmRNA在HE染色阴性淋巴结中阳性表达率为 3 9% ,在乳癌组织、HE阳性淋巴节及正常乳腺组织中阳性表达率分别为 86%、80 %及 0。结论 GST -π在肿瘤启动和促进阶段可表达 ,HE阴性淋巴结中GST -πmRNA的阳性表达表明可能存在微转移现象 ,且在耐药反应性上淋巴结的灵敏度强于乳癌组织。表明淋巴结中GST -πmRNA的表达可能是预后不良的标志 ,具有一定的临床应用价值。
Objective To investigate the correlation of placental glutathione S-transferase (GST-π) mRNA expression with breast cancer development and drug resistance in breast tumors. Methods RT-PCR technique was used to analyze the expression of GST-πmRNA in 108 negative HE stained lymph nodes from 20 breast cancer patients. Results The positive expression rate of GST-πmRNA in HE-negative lymph nodes was 39%. The positive expression rates of GST-πmRNA in breast cancer tissues, HE-positive lymph nodes, and normal breast tissues were 86%, 80%, and 0, respectively. Conclusion GST-π can be expressed during tumor initiation and promotion. The positive expression of GST-π mRNA in HE-negative lymph nodes indicates that there may be micro-metastasis, and the sensitivity of lymph node in drug-resistance is stronger than that of breast cancer. It suggests that the expression of GST-πmRNA in lymph nodes may be a sign of poor prognosis and has certain clinical application value.
