目的 探查脑梗塞灶周围区GAP-43表达的结构和时间.方法 运用电凝阻断大脑中动脉脑局部缺血模型和免疫组化(ABC)染色方法,对38只SD大鼠进行了研究.结果 GAP-43阳性反应发生于脑梗塞灶周围区的神经元的胞体和突起.其表达高峰在脑梗塞3~7d,随后逐渐减弱.神经元突起的阳性反应显示不同的形式和时相,其反应部位邻近梗塞区,在脑缺血2~3w,其显示强反应,没有发现胶质细胞的阳性反应.结论 局灶性脑缺血诱导GA-43蛋白质在梗塞灶周围区神经元表达,提示缺血区周边皮质神经元有生长和修复反应倾向.
Objective To investigate the structure and time of GAP-43 expression in the peri-infarct region of cerebral infarction.Methods Thirty-eight Sprague-Dawley rats were studied by blocking the middle cerebral artery occlusion (MCAO) model and staining by immunohistochemistry (ABC). Results The positive reaction of GAP-43 occurred in the soma and protuberance of neurons in the peripheral region of cerebral infarction.The peak of expression was gradually decreased from 3 to 7 days after cerebral infarction, then decreased gradually.The positive reaction of neurite outgrowth showed different forms and phases The reaction site was close to the infarction area, which showed strong reaction at 2 ~ 3w after cerebral ischemia, and no positive reaction was found.Conclusion Focal cerebral ischemia induces the expression of GA-43 protein in the neurons around the infarct area, suggesting that Peripheral cortical neurons in the ischemic area have a tendency to grow and repair.
