目的 探讨SLE患者外周血单个核细胞 (PBMC)凋亡率异常是否与其自身抗体产生高低有关。方法 采用碘化丙锭 (PI)染色法 ,在流式细胞仪下定量检测 2 0例SLE患者及 10名正常人PBMC体外凋亡情况 ,同时观察了SLE患者自身抗体的表达及与凋亡率的关系。结果 SLE患者PBMC体外培养 0、2 4、48、72h后凋亡率较正常人明显增高 (P <0 .0 5 ) ,且活动期SLE高于非活动期 (P <0 .0 5 )。SLE患者自身抗体产生越多者较自身抗体产生少者的凋亡率明显增高 (P <0 .0 1)。结论 SLE患者PBMC的凋亡加速 ,与其自身抗体产生增高有关 ,可能在SLE的发病机制中起一定作用
Objective To investigate whether the abnormal apoptosis rate of peripheral blood mononuclear cells (PBMC) in patients with SLE is related to the level of autoantibodies. Methods Propidium iodide (PI) staining was used to detect the apoptosis of peripheral blood mononuclear cells (PBMCs) in 20 SLE patients and 10 normal persons by flow cytometry. The autoantibodies expression and apoptosis rate in SLE patients were also observed Relationship. Results The apoptotic rates of PBMCs in SLE patients after 0,2,48 and 72h were significantly higher than those in normal controls (P <0.05), and the activity of SLE was higher than inactive (P <0. 05). SLE patients with autoantibodies produced more autoantibodies than those with less apoptosis rate was significantly increased (P <0.01). Conclusion The accelerated apoptosis of PBMC in patients with SLE may be related to the increased production of autoantibodies and may play a role in the pathogenesis of SLE
