目的:探讨五味子乙素(SchB)与5-氟尿嘧啶(5-FU)体外协同作用及对人胃癌细胞增殖及凋亡的影响。方法:MTT法检测不同浓度SchB单用或与5-FU联用对细胞增殖的抑制作用;光学显微镜观察细胞生长状态变化并计数细胞分裂指数;电镜观察药物对细胞超微结构的影响;TUNEL法检测细胞凋亡。结果:SchB单独应用对细胞增殖的抑制作用弱,与5-FU联用抑制作用显著,呈剂量依赖性;药物处理48h后,联合用药组细胞分裂指数为(1.1±0.28)%,与SchB组(13.7±0.43)%、5-FU组(5.1±0.52)%比较显著降低,P<0.05;电镜下可见细胞皱缩,胞膜界限模糊,核膜消失,核质浓缩及凋亡小体形成,以双药联用最为显著。TUNEL法分析示,联合用药组细胞凋亡率为(73.9±5.1)%,与SchB组(16.4±2.4)%、5-FU组(34.6±3.2)%比较差异有统计学意义,P<0.05。结论:SchB与5-FU联合应用能有效抑制胃癌细胞增殖,促进细胞凋亡,二药具有协同增效作用。
AIM: To investigate the synergistic effects of Schisandrin B (5-FU) and SchbB (5-FU) on the proliferation and apoptosis of human gastric cancer cells in vitro. Methods: MTT method was used to detect the inhibitory effect of Schb alone or in combination with 5-FU on cell proliferation. The changes of cell growth were observed by optical microscopy and the cell division index was counted. The effects of drugs on cell ultrastructure were observed by electron microscopy. Apoptosis was detected. Results: The inhibitory effect of SchB alone on cell proliferation was weak and was inhibited in a dose-dependent manner in combination with 5-FU. After 48 h treatment, the cell division index was (1.1 ± 0.28)% in the combination group, (13.7 ± 0.43)% in the 5-FU group (5.1 ± 0.52)%, P <0.05; under the electron microscope, cell shrinkage, blurring of the membrane boundary, disappearance of the nuclear membrane, condensation of nuclei and formation of apoptotic bodies , The most significant dual drug combination. TUNEL analysis showed that the apoptosis rate of the combination group was (73.9 ± 5.1)%, which was significantly lower than that of the Schb group (34.4 ± 3.2)% and the Schb group (16.4 ± 2.4)%, P <0.05 . Conclusion: The combined use of SchB and 5-FU can effectively inhibit the proliferation of gastric cancer cells and promote apoptosis, and the two drugs have synergistic effects.
