目的应用免疫速率比浊方法测定A群流行性脑膜炎球菌多糖含量,并进行验证。方法采用免疫速率比浊方法测定A群脑膜炎球菌多糖标准品多糖含量,绘制标准曲线,对该方法的精密性、准确性、灵敏性进行验证,并进行干扰试验。结果多糖抗原浓度在0~10μg/ml之间,与相应的浊度值呈良好的线性关系,R2=0.984 3;该方法检测结果的日内和日间CV值均小于5%,不同浓度A-DT结合物样品的回收率在99.67%~102.00%之间,最低检出限为0.159μg/ml;加入C群流行性脑膜炎球菌多糖后的A群脑膜炎球菌多糖含量与未添加相比,差异无统计学意义(P>0.05)。结论免疫速率比浊方法测定A群脑膜炎球菌多糖含量具有较高的准确度、精确度及灵敏度,并具有较强的特异性。
Objective To determine the epidemic meningococcal polysaccharide content of group A by immunoturbidimetry and verify the results. Methods The polysaccharide content of group A meningococcal polysaccharide was determined by immunoturbidimetry. The standard curve was drawn and the precision, accuracy and sensitivity of the method were verified and the interference test was carried out. Results The polysaccharide antigen concentration was between 0 and 10 μg / ml, with a good linear relationship with the corresponding turbidity value, R2 = 0.984 3. The intra-and inter-day CV values of the method were less than 5% The recoveries of DT conjugate samples ranged from 99.67% to 102.00% with the minimum detectable limit of 0.159μg / ml. The meningococcal group A polysaccharide content of group C meningococcal polysaccharide was significantly higher than that without addition of meningococcal group C, The difference was not statistically significant (P> 0.05). Conclusion The immunoturbidimetric method has high accuracy, accuracy and sensitivity for the determination of the meningococcal polysaccharide group A and has strong specificity.
