目的探讨大肠杆菌疫苗对发育期胎兔肺表面活性物质磷脂生成的影响。方法将孕兔30只随机分为3组,大肠杆菌疫苗组10只,地塞米松组10只,生理盐水组10只。大肠杆菌疫苗组和生理盐水组分别于妊娠第10、17、24天宫颈肌肉内注射大肠杆菌疫苗0.1 ml/kg和生理盐水0.1 ml/kg,地塞米松组于妊娠第22、23、24天分别肌肉注射地塞米松0.4 ml/kg。于妊振第27天剖宫产,行支气管肺泡灌洗收集管肺泡灌洗液(BALF),采用高效液相色谱-蒸发光散射法检测BALF中二棕榈磷脂酰胆碱(DPPC)的浓度,采用二喹啉甲酸(BCA)法测定BALF上清液总蛋白浓度,采用动态定时散射比浊法测定羊水和BALF中CRP浓度;采用免疫荧光法测定羊水和BALF中PCT浓度。采用单因素方差分析;LSD和Duunet T3法进行统计学分析。结果地塞米松组、大肠杆菌疫苗组、生理盐水组胎肺BALF中DPPC的浓度分别为(44.89±0.29)μg/ml、(44.62±0.11)μg/ml、(40.20±0.27)μg/ml,地塞米松组和大肠杆菌疫苗组胎肺BALF中DPPC的浓度明显高于生理盐水组,差异有统计学意义(P<0.01);大肠杆菌疫苗组、地塞米松组、生理盐水组BALF上清液总蛋白浓度分别为(0.409±0.012)μg/ml、(0.253±0.043)μg/ml、(0.244±0.019)μg/ml,大肠杆菌疫苗组BALF上清液总蛋白浓度高于地塞米松组和生理盐水组,差异有统计学意义(P<0.01);大肠杆菌疫苗组羊水和BALF中CRP、PCT明显高于地塞米松组和生理盐水组,差异有统计学意义(P<0.01)。结论大肠杆菌疫苗诱导宫内感染和产前注射地塞米松对胎兔肺DPPC生成均有明显的促进作用,接种大肠杆菌疫苗可能对胎肺上皮细胞造成一定程度的损伤。
Objective To investigate the effect of Escherichia coli (E.coli) vaccine on the production of pulmonary surfactant in developing rabbit. Methods Thirty pregnant rabbits were randomly divided into three groups: 10 in E. coli vaccine group, 10 in dexamethasone group and 10 in saline group. Escherichia coli vaccine group and saline group were injected intraperitoneally with E.coli vaccine 0.1 ml / kg and saline 0.1 ml / kg respectively on the 10th, 17th and 24th day of gestation, and dexamethasone group at the 22th, 23th and 24th day of gestation Dexamethasone 0.4 ml / kg were injected intramuscularly. BALF was collected in bronchoalveolar lavage fluid (BALF) on the 27th day of pregnancy and then the concentration of dipalmitoylphosphatidylcholine (DPPC) in BALF was measured by high performance liquid chromatography coupled with evaporative light scattering. The total protein concentration of BALF supernatant was determined by bicinchoninic acid (BCA) method. The concentration of CRP in amniotic fluid and BALF was determined by dynamic time-of-flight turbidimetry. The PCT concentration in amniotic fluid and BALF was measured by immunofluorescence. One-way ANOVA and LSD and Duunet T3 were used for statistical analysis. Results The concentrations of DPPC in fetal lung and BALF in dexamethasone group, E. coli vaccine group and saline group were (44.89 ± 0.29) μg / ml, (44.62 ± 0.11) μg / ml and (40.20 ± 0.27) μg / The concentration of DPPC in fetal lung and BALF of dexamethasone and Escherichia coli vaccine groups was significantly higher than that of saline group (P <0.01).
