目的 运用cDNA微阵列技术研究口腔白斑组织和口腔鳞癌组织的基因表达谱的变化 ,为探讨口腔白斑癌变基因提供初步筛选资料。方法 对临床切除的 5例口腔白斑和口腔癌组织进行总RNA抽提 ,逆转录制备探针 ,纯化后与含有 412 4个基因的微阵列杂交 ,杂交后的信号经扫描仪检测和计算机分析后筛选出与白斑癌变相关的基因。结果 对口腔白斑和口腔癌基因表达谱进行分析 ,发现存在 3 0个差异表达的基因 ,它们可能与白斑癌变有关。结论 采用cDNA微阵列技术能成功检测出两种不同组织中多个基因的差异表达 ,为研究白斑癌变机制提供有价值的筛选资料
Objective To study the changes of gene expression profile in oral leukoplakia and oral squamous cell carcinoma using cDNA microarray technology and to provide preliminary screening data for exploring the gene of oral leukosis. Methods Total RNA was extracted from 5 cases of clinically isolated oral leukoplakia and oral cancer tissues. The probe was reverse transcribed to prepare the probe. After hybridization, it was hybridized with a microarray containing 4124 genes. After hybridization, the signal was detected by scanner and analyzed by computer Screening out the genes associated with leukoplakia. Results Oral leukoplakia and oral cancer gene expression profiles were analyzed and found there were 30 differentially expressed genes, which may be related to leukoplakia. Conclusion The differential expression of multiple genes in two different tissues can be successfully detected by cDNA microarray, which provides valuable screening data for studying the carcinogenesis of leukoplakia
