为克服从小麦叶片提取DNA用于PCR受季节的限制以及氯仿不易购买等问题,研究以从叶片中提取的以及用氯仿从小麦籽粒中提取的DNA为对照,用无氯仿方法提取小麦籽粒中的DNA,并用多重PCR进行检测。结果表明,用无氯仿方法提取的DNA浓度比从叶片中提取的和用氯仿提取的DNA浓度要低,而且用无氯仿方法提取的DNA扩增效果稍差,但该问题可通过增加上样量或采用聚丙稀酰胺凝胶电泳等途径来解决。因此,无氯仿小麦籽粒DNA提取方法可在对DNA质量要求不高以及氯仿无法及时得到时使用。
In order to overcome the problem of DNA extraction from wheat leaves for seasonal restriction of PCR and the difficulty of chloroform purchase, the extraction of DNA from leaves and extraction of DNA from wheat seeds with chloroform was used as a control, chloroform extraction was used to extract DNA and tested by multiplex PCR. The results showed that the concentration of DNA extracted by chloroform-free method was lower than the concentration of DNA extracted from leaves and extracted with chloroform, and the DNA extracted by chloroform-free method was slightly less effective. However, this problem could be solved by increasing the loading amount Or using polyacrylamide gel electrophoresis and other means to solve. Therefore, the DNA extraction method of chloroform-free wheat seeds can be used when the quality of DNA is not high enough and chloroform can not be obtained in time.
