目的探讨食管鳞状细胞癌中基质金属蛋白酶-8(matrix metalloproteinase 8,MMP-8)的表达及其与肿瘤浸润、转移的关系。方法实时荧光定量PCR方法检测67例食管鳞癌组织和27例远癌组织中MMP-8 mRNA的表达;Western blot法检测其中15例食管鳞癌组织及其配对的15例远癌组织中MMP-8蛋白的表达,统计分析MMP-8表达水平与食管鳞癌的临床分期以及浸润转移的关系。结果①MMP-8在食管鳞癌组织和远癌组织中均有不同程度的表达,在癌组织的表达水平显著低于远癌组织(mRNA表达水平P=0.003,蛋白表达水平P=0.031)。②T1、T2、T3、T4期食管鳞癌组织中MMP-8 mRNA表达存在显著差异(P=0.002),MMP-8表达随着肿瘤浸润程度的加深而升高;MMP-8蛋白表达量在T1~T4期中也呈增加的趋势。③MMP-8 mRNA和蛋白表达水平在食管鳞癌的不同淋巴结转移组之间没有显著差异。结论 MMP-8在食管鳞癌组织中低表达,但又随肿瘤浸润加深而表达升高,可能反映了MMP-8在食管鳞癌发生和浸润中的不同作用。
Objective To investigate the expression of matrix metalloproteinase-8 (MMP-8) in esophageal squamous cell carcinoma and its relationship with tumor invasion and metastasis. Methods The expressions of MMP-8 mRNA in 67 esophageal squamous cell carcinoma and 27 distant esophageal squamous cell carcinoma tissues were detected by real-time fluorescence quantitative PCR. The expression of MMP-8 mRNA in 15 esophageal squamous cell carcinoma tissues and 15 matched esophageal squamous cell carcinoma tissues was detected by Western blot. 8 protein expression, statistical analysis of MMP-8 expression and esophageal squamous cell carcinoma of the clinical stage and the relationship between invasion and metastasis. Results ①MMP-8 was expressed in esophageal squamous cell carcinoma and distant-cancer tissues to varying degrees. The expression level of MMP-8 in cancer tissues was significantly lower than that in distant cancer tissues (P = 0.003, P = 0.031). The expression of MMP-8 mRNA in T1, T2, T3 and T4 esophageal squamous cell carcinoma tissues was significantly different (P = 0.002), while the expression of MMP-8 increased with the progression of tumor invasion. The expression of MMP-8 protein in T1 ~ T4 period also showed an increasing trend. ③MMP-8 mRNA and protein expression levels in esophageal squamous cell carcinoma of different lymph node metastasis group did not differ significantly. Conclusions The low expression of MMP-8 in esophageal squamous cell carcinoma, but with the deepening of tumor invasion and expression, may reflect the different roles of MMP-8 in the occurrence and invasion of esophageal squamous cell carcinoma.
