以小球藻为原料,分离出色素蛋白复合体,并对其组成及抗氧化性质进行表征。用超声波破碎小球藻,收集类囊体膜后,加入两性离子表面活性剂CHAPS增溶,得到小球藻色素蛋白复合体。对色素蛋白复合体的组成进行分析,用HPLC法测定叶黄素含量,分光光度法测定叶绿素含量及BCA法测定蛋白质含量,结果表明:复合体中m叶黄素∶m叶绿素∶m蛋白=12.49∶7.18∶100。分离得到的小球藻色素蛋白复合体对DPPH自由基、超氧阴离子自由基的半数清除质量浓度分别为422μg/m L和294μg/m L;当其质量浓度为1 000μg/m L时,金属螯合率达30%;还原力随色素蛋白复合体质量浓度的上升而增强;复合体质量浓度600μg/m L时,可完全抑制脂质过氧化。
Using chlorella as raw material, the pigment protein complex was isolated, and its composition and anti-oxidation properties were characterized. After breaking the chlorella by ultrasonic wave and collecting the thylakoid membrane, the zwitterionic surfactant CHAPS is added to obtain the chlorella pigment protein complex. The composition of the pigment protein complex was analyzed, the content of lutein was determined by HPLC, the content of chlorophyll was determined by spectrophotometry and the protein content by BCA method. The results showed that the content of m-lutein: m chlorophyll: m protein was 12.49 : 7.18: 100. The half-masses of DPPH free radical and superoxide anion free radicals were respectively 422μg / mL and 294μg / mL when the mass concentration of Chlorella chromogen protein complex was 1 000μg / m L, The chelating rate was 30%. The reducing power increased with the increase of the mass concentration of the pigment protein complex. When the complex concentration was 600 μg / mL, lipid peroxidation could be completely inhibited.
