目的探讨清脑止痛胶囊(QNZTJN)对硝酸甘油致偏头痛大鼠的影响。方法健康雄性Wistar大鼠90只,随机分为6组:空白组、模型组、清脑止痛胶囊低(QNZTJN-L,0.35g/kg)、中(QNZTJN-M,0.7g/kg)、高(QNZTJN-H,1.4g/kg)剂量组、阳性药正天丸组(0.006g/kg),每组15只。给药7天后,用皮下注射硝酸甘油(NTG,10mg/kg)复制大鼠偏头痛模型,观察注射硝酸甘油后180min内大鼠行为学反应,造模4h后检测血清及脑干中一氧化氮(NO)、一氧化氮合酶(NOS)、5-羟色胺(5-HT)、降钙素基因相关肽(CGRP)、β-内啡肽(β-EP)含量的变化。结果与对照组比较,模型组大鼠挠头次数明显增加(P<0.05,P<0.01),血浆和脑组织NO含量明显升高(P<0.05,P<0.01),脑组织中NOS、CGRP含量明显升高(P<0.05),β-EP、5-HT含量明显降低(P<0.05);与模型组比较,清脑止痛胶囊中、高剂量组大鼠挠头次数明显减少(P<0.05,P<0.01),血浆和脑组织NO含量明显降低(P<0.05,P<0.01),脑组织中NOS、CGRP含量明显降低(P<0.05),β-EP、5-HT含量明显升高(P<0.05),且呈剂量依赖性。结论清脑止痛胶囊可以缓解大鼠偏头痛症状,其机制可能与调节单胺类神经递质释放,进而改善脑及血管的功能障碍有关。
Objective To investigate the effect of qingnaozhitong capsule (QNZTJN) on nitroglycerin-induced migraine in rats. Methods Ninety male Wistar rats were randomly divided into 6 groups: blank group, model group, QNZTJN-L (0.35g / kg), QNZTJN-M (QNZTJN-H, 1.4g / kg) dose group, positive drug Zhengtian pill group (0.006g / kg), 15 rats in each group. Seven days after administration, nitroglycerin (NTG, 10 mg / kg) was subcutaneously injected into the migraine model to observe the behavioral response within 180 min after injection of nitroglycerin. Compared with the model group, the number of scratching in the middle and high dose Qingnaizhengtong capsules group was significantly decreased (P <0.05, P < (P <0.01, P <0.01). The contents of NO and NO in plasma and brain tissues were significantly decreased (P <0.05, P <0.01) P <0.05), and in a dose-dependent manner. Conclusions Qingnaozhitong capsule can relieve migraine symptoms in rats. The mechanism may be related to the release of monoamine neurotransmitter and the improvement of brain and blood vessel dysfunction.
