半枝莲总黄酮对ApoE基因敲除小鼠血脂及抗氧化功能的影响

目的探讨半枝莲总黄酮对载脂蛋白E基因敲除小鼠动脉粥样硬化病变形成早期血清脂质水平、丙二醛(MDA)、超氧化物歧化酶(SOD)表达的影响。方法载脂蛋白E基因敲除小鼠随机分为模型组、辛伐他汀组、银杏叶胶囊组和半枝莲总黄酮高、中、低剂量组,取正常C57BL/6小鼠为对照组,每组6只。除模型组、对照组给予等容量羧甲基纤维素钠外,其他各组分别给予阳性对照药与受试药8周。处死小鼠,ELISA法检测血脂水平,黄嘌呤氧化酶法检测SOD活力,硫代巴比妥酸法测定MDA含量,HE染色观察主动脉形态学变化。结果半枝莲总黄酮中、高剂量组血清甘油三酯(TG)、总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)水平明显低于模型组,高剂量组高密度脂蛋白胆固醇(HDL-C)水平明显高于模型组(P<0.05或0.01)。半枝莲总黄酮3个剂量组的MDA含量较模型组显著减少,SOD活性表达水平较模型组显著升高(均P<0.01)。结论半枝莲总黄酮可通过降低载脂蛋白E基因敲除小鼠TG、TC、LDL-C水平,升高HDL-C水平,升高SOD活性,降低MDA含量,从而发挥抗动脉粥样硬化的作用。 Objective To investigate the effects of total flavonoids of Scutellaria barbata on serum lipid levels, malondialdehyde (MDA) and superoxide dismutase (SOD) in the early stage of atherosclerosis induced by apolipoprotein E gene knockout mice. Methods Apolipoprotein E gene knockout mice were randomly divided into model group, simvastatin group, Ginkgo biloba capsule group and Scutellaria barbata total flavonoids high, medium and low dose groups, normal C57BL / 6 mice as control group, 6 per group In addition to the model group, the control group given equal volume of sodium carboxymethyl cellulose, the other groups were given positive control drug and test drug for 8 weeks. Mice were sacrificed, serum lipid levels were measured by ELISA, SOD activity was measured by xanthine oxidase method, MDA content was measured by thiobarbituric acid method, morphological changes of aorta were observed by HE staining. Results The levels of triglyceride (TG), total cholesterol (TC) and low density lipoprotein cholesterol (LDL-C) in Scutellaria barbata total flavonoids were significantly lower than those in model group and high-dose high-density lipoprotein cholesterol (HDL-C) was significantly higher than the model group (P <0.05 or 0.01). Scutellaria barbata total flavonoids in the three doses of MDA content than the model group decreased significantly, SOD activity was significantly higher than the model group (P <0.01). Conclusion Scutellaria barbata total flavonoids can reduce the levels of TG, TC and LDL-C in the apolipoprotein E knockout mice, increase the level of HDL-C, increase the activity of SOD and decrease the content of MDA, thus play an important role in anti-atherosclerosis Role.
