目的研究艾滋病病毒(HIV)感染者唾液酸结合性免疫球蛋白样凝集素7(Siglec-7~+)自然杀伤(NK)细胞亚群数量和功能的变化,探讨Siglec-7~+NK细胞亚群数量下降的原因,为临床治疗提供免疫学理论基础。方法(1)利用流式细胞学技术检测Siglec-7~+NK细胞亚群的数量和凋亡相关因子(FAS/CD95)的表达情况;(2)白介素12/18(IL-12/18)刺激培养后,观察不同NK细胞亚群的功能。结果 HIV感染组NK细胞数量减少,尤其是Siglec-7~+ CD56~(dim)NK细胞群亚群下降更显著(P<0.01)。功能方面,通过IL-12/18的刺激,无论在健康人组还是HIV感染组,Siglec-7~+NK细胞亚群分泌γ干扰素(IFN-γ)的能力明显强于阴性亚群,但在HIV感染组NK细胞分泌IFN-γ功能下降,低于健康人组(P<0.001);同时Siglec-7~+ CD56~(dim)NK细胞亚群FAS的表达在HIV感染组明显高于健康人组(P<0.05),且Siglec-7~+ CD56~(dim)NK细胞的数量与CD95的表达呈负相关(r=—0.472,P<0.01)。结论 Siglec-7~+NK细胞亚群功能优于阴性亚群,但在HIV感染中本群发生明显下降,Siglec-7~+ CD56~(dim)NK细胞亚群数量与CD95的表达呈负相关。
Objective To study the changes of natural killer (NK) cell subsets and function of sialic acid-binding immunoglobulin-like agglutinin-7 (+) in HIV-infected patients and explore the role of Siglec-7 ~ + NK cells The reasons for the decline in the number of groups provide clinical basis for immunological theory. Methods (1) The number of Siglec-7 ~ + NK cell subsets and the expression of apoptosis related factor (FAS / CD95) were detected by flow cytometry; (2) The levels of interleukin 12/18 After stimulating the culture, the function of different NK cell subsets was observed. Results The number of NK cells in HIV-infected group decreased, especially in Siglec-7 ~ + CD56-dim NK cell population decreased more significantly (P <0.01). Functionally, Siglec-7 ~ + NK cell subsets secreted interferon-γ (IFN-γ) were significantly stronger than negative ones in IL-12/18 stimulated IL-12/18 cells both in healthy and HIV-infected groups The IFN-γ secretion of NK cells in HIV-infected group was lower than that in healthy people (P <0.001). Meanwhile, the expression of FAS in Siglec-7 ~ + CD56-dim cell subsets was significantly higher in HIV-infected patients than in healthy controls (P <0.05). There was a negative correlation between the number of Siglec-7 + CD56D NK cells and CD95 expression (r = -0.472, P <0.01). Conclusion The function of Siglec-7 ~ + NK cell subsets is better than that of negative subgroups, but the number of Siglec-7 ~ + CD56 ~ (+) diminished NK cell subsets is negatively correlated with the expression of CD95 .
