目的:观察痹肿消汤对胶原诱导性关节炎大鼠滑膜AnnexinⅠ、AnnexinⅤ表达的影响,探讨痹肿消汤治疗RA的作用机制。方法:70只SD大鼠随机分为3组,皮下注射牛Ⅱ型胶原与完全弗氏佐刺诱导SD大鼠实验性关节炎,Western blotting方法检测各组不同时间点大鼠滑膜组织AnnexinⅠ、AnnexinⅤ的表达。结果:造模大鼠88%出现关节炎症状;造模25天后,模型组AnnexinⅠ、AnnexinⅤ表达水平明显低于正常组(P<0.05);痹肿消汤组AnnexinⅠ、AnnexinⅤ表达高于模型组(P<0.01);随着时间延长,模型组AnnexinⅠ、Annexin水平逐渐降低,痹肿消汤组则逐渐增强。结论:AnnexinⅠ、Annexin在RA滑膜组织炎症形成和发展中发挥着重要作用;痹肿消汤通过上调滑膜AnnexinⅠ和Annexin表达水平发挥作用。
OBJECTIVE: To observe the effect of Qizhongxiao Decoction on the expression of Annexin I and AnnexinV in synovial membrane of collagen-induced arthritis in rats, and to explore the mechanism of the treatment of RA with Qixiaxiao Decoction. Methods: Seventy Sprague-Dawley rats were randomly divided into 3 groups. Experimental arthritis was induced in SD rats by subcutaneous injection of bovine type II collagen and complete Freunds koji injection. Western blotting was used to detect the synovial tissue Annexin I in each group. AnnexinV expression. RESULTS: Arthritis symptoms occurred in 88% of the model rats; after 25 days of modeling, the expression levels of Annexin I and AnnexinV in the model group were significantly lower than those in the normal group (P<0.05); the expression of Annexin I and AnnexinV in the QFXD group was higher than that in the model group. Group (P <0.01); As time went on, the levels of Annexin I and Annexin in the model group gradually decreased, while the Biaozhongxiao Decoction group gradually increased. Conclusion: Annexin I and Annexin play an important role in the formation and development of inflammation in RA synovial tissue. Bianzhongxiao Decoction plays a role by up-regulating the expression levels of Annexin I and Annexin in synovium.
