通过分子标记辅助选择,将稻瘟病抗性材料BL108的抗性基因Pi25导入光温敏核不育系浙科22S,利用Pi25紧密连锁STS标记Si13070D检测目的基因,在浙科22S/BL108 F3株系中,获得39份呈Pi25纯合基因型不育系。经稻瘟病混合菌种接种鉴定,有29个株系表现抗病,与分子标记辅助选择的准确率为85.23%。结果表明,STS标记Si13070D是检测Pi25基因的有效分子标记。在分子检测和抗性表现的基础上,根据育性、株叶形态等性状,获得了5个综合性状表现良好的两系不育系16S、38S、39S、61S和73S。
The molecular marker-assisted selection was used to transfer the resistance gene Pi25 of rice blast resistance material BL108 into Zhenshu 22S which is a photo-thermo-sensitive genic male sterile line. The target gene was detected by Pi25 close-linkage STS-labeled Si13070D. , 39 Pi25 homozygous genotype male sterile lines were obtained. Inoculation identification of mixed blast of rice blast, 29 lines showed disease resistance, and the accuracy of molecular marker-assisted selection was 85.23%. The results showed that the STS marker Si13070D is an effective molecular marker for detecting Pi25 gene. Based on the molecular detection and resistance performance, five lines of CMS 16S, 38S, 39S, 61S and 73S were obtained according to the characteristics of fertility and plant morphology.
