目的:研究肝硬化大鼠肝脏、脾脏血红素氧化酶(HO-1)、HO-2的定位表达的改变。方法:制备大鼠肝硬化模型,用免疫组化染色法观察HO-1、HO-2在细胞内的分布,并在显微镜下用计算机图像分析技术计算两者表达的强弱。结果:与对照组相比,肝硬化组肝脏、脾脏、门静脉HO-1阳性细胞的表达显著增加,且主要分布在肝脏的门管区周围及脾脏的红髓区,而HO-2的变化不明显。结论:HO可能参与肝硬化的发生过程、与门脉高压血流动力学紊乱有关。
Objective: To study the changes of the localization of heme oxygenase (HO-1) and HO-2 in liver and spleen of cirrhotic rats. Methods: The rat model of liver cirrhosis was established. The distribution of HO-1 and HO-2 in the cells was observed by immunohistochemistry. The expression of HO-1 and HO-2 in the cells was analyzed under a microscope by computer image analysis. Results: Compared with the control group, the expression of HO-1 positive cells in liver, spleen and portal vein of cirrhotic patients increased significantly, and mainly distributed around the portal area of the liver and the red marrow of spleen, while the change of HO-2 was not obvious . Conclusion: HO may be involved in the pathogenesis of liver cirrhosis, which is related to the disturbance of portal hemodynamics.
