雷公藤多甙(GTW)在雄性大鼠及男子均可致不育而无明显毒副作用。本实验用BABL/C小鼠骨髓细胞体内Brdu标记姐妹染色单体互换(SCE)的方法,测定GTW有无潜在的致突变作用。采用临床剂量3倍(3mg/kg)和100倍(100mg/kg),每天灌服,共 7天。另设阳性对照组一次性腹腔注射环磷酰胺30mg/kg。结果,SCE率范围低剂量组在1~7之间,平均值3.63±0.20(SE),高剂量组在1~15之间,平均值 4.56±0.39,阴性对照组在1~8之间,平均值 3.8±0.39,阳性对照组在 19~58之间,平均值33.1±1.06。经统计学处理,低、高剂量与阴性对照组之间无明显差异,而阳性对照组和其他各组之间均有显著性差异(P<0.01)。因而从本实验的结果来看,GTW似无诱变作用。
Tripterygium wilfordii (GTW) can cause infertility in male rats and men without obvious side effects. In this experiment, BADH/C mouse bone marrow cells in vivo Brdu-labeled sister chromatid exchange (SCE) method to determine the potential mutagenic effect of GTW. Three doses (3 mg/kg) and 100 (100 mg/kg) of the clinical dose were given daily for 7 days. A separate positive control group was given by intraperitoneal injection of cyclophosphamide 30 mg/kg. As a result, the low-dose SCE range was between 1 and 7, with an average of 3.63±0.20 (SE), between 1 and 15 in the high-dose group, with an average of 4.56±0.39, and between 1 and 8 in the negative control group. The mean value was 3.8±0.39. The positive control group was between 19 and 58 with an average of 33.1±1.06. After statistical analysis, there was no significant difference between the low and high doses and the negative control group, but there was a significant difference between the positive control group and other groups (P<0.01). Therefore, from the results of this experiment, GTW seems to have no mutagenic effect.
