为探明烟用底料变质的原因,对底料的微生物种群、菌落数量及致香成分进行了分析.利用分离培养基从正常紫云底料、变质紫云底料、正常软珍底料、变质软珍底料中分离、鉴定细菌和真菌,得到28株细菌和1株真菌.真菌仅有酵母菌属(Saccharomyces sp.),细菌分属于12个属,包括芽孢杆菌属(Bacillus sp.)、微杆菌属(Microbacterium sp.)、鞘氨醇单胞菌属(Sphingomonas sp.)、勒克菌属(Leclercia sp.)、不动杆菌属(Acinetobacter sp.)、短波单胞菌属(Brevundimonas sp.)、土壤杆菌属(Agrobacterium sp.)、青枯菌属(Ralstonia sp.)、醋酸杆菌属(Acetobacterium sp.)、燕麦菌属(Acidovorax sp.)、类鼻疽菌属(Burkholderia sp.)、肠杆菌属(Enterobacter sp.).变质紫云底料中微生物菌落总数是正常紫云底料微生物菌落总数的90倍,变质软珍底料微生物菌落总数是正常软珍底料微生物菌落总数的29倍.对微生物菌落总数的动态变化进行了追踪调查,结果表明,烟用底料在室温条件下6d即可变质.利用气相色谱-质谱联用仪对正常和变质底料致香成分及含量(质量分数)进行检测,发现变质后的底料致香成分及含量变化较大.在紫云底料中β-二氢大马酮、δ-二氢大马酮含量变化最大,薄荷醇在变质紫云底料中没有检测到;软珍底料中辛酸、癸酸、癸酸乙酯、δ-二氢大马酮、β-二氢大马酮含量变化最大,辛酸乙酯、薄荷醇、烟酮、苯甲醛、α-二氢大马酮在变质软珍底料中没有检测到.“,”To investigate possible reasons behind the deterioration of base casing for tobacco, tobacco microbial population, colony quantity and aroma components were analyzed. Using common separation medium, 28 bacteria and 1 fungus were isolated and identified from the normal and deteriorated base casings for cigarette brands“Purple Yun Yan”and“Soft Precious Yun Yan”. The fungus belonged to Saccharomyces sp. and 28 bacteria belonged to 12 genera, including Bacillus sp., Microbacterium sp., Sphingomonas sp., Leclercia sp.,Acinetobacter sp., Brevundimonas sp., Agrobacterium sp., Ralstonia sp., Acetobacterium sp., Acidovorax sp., Burkholderia sp., Enterobacter sp. The total microorganism populations in the two deteriorated base casings were 90 and 29 times of those in the normal casings for the same brands, respectively. The dynamic variations of total microorganism population were investigated, the results showed that base casing started to deteriorate from the 6th day at room temperature. The types and contents of aroma components in normal and deteriorated base casings were detected by two-dimensional gas chromatography-mass spectrometry (GC-MS) and the results indicated that the types and contents changed greatly after deterioration. In the deteriorated base casing for cigarette brand“Purple Yun Yan”, the contents of β-damascone and δ-damascone changed most significantly, while menthol was not detectable. In the deteriorated base casing for cigarette brand“Soft Precious Yun Yan”, the contents of octanoic acid, decyl acid, ethyl ester, β-damascone and δ-damascone changed most significantly, while octanoic acid ethyl ester, menthol, 3,5,5-trimethylcyclohexane-1,2-dione, benzaldehyde, α-damascone were not detectable.
