本文报道用具化学发光检测的HPLC(HPLCCL)系统,可以测定体液中的吗啡.实验采用Sep Pak,C_(13)柱和Bond-Elut C_(18)柱分别对尿及血样进行预处理,分离全血和尿中吗啡,用HPLC-CL测定其含量.仪器用HPLC Perkin-Elmer 3B,不锈钢柱为15cm×4.6mm(id),内装8μm反相苯乙烯-二乙烯基苯固定相,流动相为0.01mol/L聚磷酸-甲醇(87.5∶12.5,v/v),流速1ml/min,氧化剂6×10~(-4)mol/LKMnO_4溶液(流速0.4ml/min),CL检测器于1160V下操作.实验结果表明,CL对HPLC柱后反应是一个理想的光谱检测技术.CL反应快速,特别适合于柱后反应检测.吗啡的CL反应是在KMnO_4存在下,酸性聚
This paper reports the determination of morphine in body fluids by HPLC (HPLC) with chemiluminescence detection.The urine and blood samples were pretreated with Sep Pak, C_ (13) and Bond-Elut C_ (18) Blood and urinary morphine were determined by HPLC-CL.The instrument used HPLC Perkin-Elmer 3B, stainless steel column 15cm × 4.6mm (id), containing 8μm reverse phase styrene-divinylbenzene stationary phase, the mobile phase was 0.01 mol / L polyphosphoric acid-methanol (87.5: 12.5, v / v), flow rate 1 ml / min and oxidant solution 6 × 10 -4 mol / L KMnO 4 at a flow rate of 0.4 ml / min. .The experimental results show that CL is an ideal spectral detection technique after HPLC column.The CL reaction is rapid and is particularly suitable for the post-column reaction detection.Morphine CL reaction is in the presence of KMnO_4 acid poly
