目的通过探索D-聚甘酯抗凝活性与Ⅹ/Ⅱ因子灭活度比值的相关性,推断其可能的抗凝抗栓机制,为临床更加合理应用该药提供参考。方法设4个不同D-聚甘酯剂量组以及空白、肝素钠2个对照组,每组均测定凝血酶原时间(PT),凝血活酶时间(APTT)以及Ⅱ因子、Ⅹ因子的活性,并计算Ⅹ/Ⅱ的灭活度比值。结果在50~150μg.mL?1内,随着D-聚甘酯浓度的增加,其抗凝活性增强,Ⅹ/Ⅱ因子灭活度比值升高。结论 D-聚甘酯在发挥抗凝活性时,可能的机制是以灭活凝血酶活性为主,而不是以抑制凝血酶原产生为主。
OBJECTIVE: To explore the possible anticoagulant and antithrombotic mechanism by exploring the correlation between the anticoagulant activity of D-polyglycerol and the ratio of factor X / II inactivation, and to provide a reference for the more rational use of this drug in clinic. Methods Four different dosages of D-polyglycerol and two control groups of blank and heparin sodium were set up. The activity of prothrombin time (PT), thromboplastin time (APTT), factor II and factor X were measured in each group. And calculate the ratio of inactivation of X / II. Results In the range of 50 ~ 150μg · mL -1, the anticoagulant activity increased and the ratio of factor X / Ⅱ inactivation increased with the increase of D-polyglycerol concentration. Conclusion The possible mechanism of D-polyglycerol to exert anticoagulant activity is to inactivate thrombin activity rather than to inhibit the production of prothrombin.
