目的本实验通过分析口腔鳞癌中SOX7基因的甲基化状态及其mRNA表达情况,初步探索口腔鳞癌的发生发展机制。方法收集河北医科大学第四医院口腔鳞癌组织及正常口腔黏膜组织标本48例,采用甲基化特异性PCR(methylation specific PCR,MSP)及RT-PCR(reverse transcrpition polymerase chain reaction)分析口腔鳞癌组织及正常口腔黏膜组织中SOX7基因甲基化状态、SOX7 mRNA表达情况与临床病理特征之间的关系。结果 OSCC组织中SOX7基因甲基化阳性率高于正常口腔黏膜(43.75%vs 20.83%,P<0.05),该基因的高甲基化状态与淋巴结转移、临床分期及病理分级相关(P<0.05)。OSCC组织中SOX7基因mRNA的表达量明显低于正常口腔黏膜(0.41±0.20 vs0.74±0.40,P<0.001),该基因的低表达与淋巴结转移、临床分期及病理分级相关(P<0.05)。同时OSCC组织中SOX7基因mRNA的表达情况与SOX7基因的甲基化状态相关(P<0.05)。结论 SOX7基因启动子区CpG岛高甲基化可能与口腔鳞癌的发生及进展相关,有可能成为口腔鳞癌评估预后的重要指标。
Objective To investigate the methylation status of SOX7 gene and its mRNA expression in oral squamous cell carcinoma (OSCC), and to explore the mechanism of the occurrence and development of OSCC. Methods Forty eight patients with oral squamous cell carcinoma and normal oral mucosa were collected from the Fourth Hospital of Hebei Medical University. Methylation-specific PCR (MSP) and reverse transcrpition polymerase chain reaction (RT-PCR) Relationship between methylation status of SOX7 gene, SOX7 mRNA expression and clinicopathological features in tissues and normal oral mucosa. Results The positive rate of methylation of SOX7 gene in OSCC tissues was significantly higher than that in normal oral mucosa (43.75% vs 20.83%, P <0.05). The hypermethylation status of this gene correlated with lymph node metastasis, clinical stage and pathological grade (P <0.05). The expression of SOX7 mRNA in OSCC was significantly lower than that in normal oral mucosa (0.41 ± 0.20 vs 0.74 ± 0.40, P <0.001). The low expression of SOX7 mRNA was correlated with lymph node metastasis, clinical stage and pathological grade (P <0.05) . Meanwhile, the expression of SOX7 mRNA in OSCC was correlated with the methylation status of SOX7 gene (P <0.05). Conclusion The hypermethylation of CpG island in promoter region of SOX7 gene may be related to the occurrence and progression of oral squamous cell carcinoma, which may be an important index for evaluating the prognosis of oral squamous cell carcinoma.
