目的:探讨胚胎发育过程中鸡肢体骨骼肌生肌因子与乙酰胆碱受体基因的时相表达及相互关系。方法:采用Northern印迹杂交检测不同发育阶段鸡胚和雏鸡肢体骨骼肌组织MyoD、生肌素、AChRα亚基、α肌动蛋白和原癌基因fos等在转录水平的表达。结果:各种因子及结构蛋白在鸡肢体骨骼肌发育过程中呈现各自独特的表达模式。结论:在鸡不同发育时期,生肌因子MyoD、生肌素,α肌动蛋白在肢体骨骼肌表达时相与躯干肌表达时相不完全相同;生肌因子与AChRα亚基的表达同受神经植人影响,发育早期AChR表达不依赖生肌素,可能受MyoD家族其它成员或另类因子的调节。
OBJECTIVE: To investigate the time-phase expression of skeletal muscle myogenic factor and acetylcholine receptor genes during chicken embryo development and their correlation. Methods: Northern blotting was used to detect the expression of MyoD, myogenin, AChRα subunit, α-actin and proto-oncogene fos in skeletal muscle of chick embryo and chickens at different developmental stages. Results: Various factors and structural proteins showed their own unique expression pattern during the development of skeletal muscle in chicken limbs. CONCLUSION: Myogenic factor MyoD, myogenin and α-actin are not completely the same in trunk skeletal muscle during the different developmental stages of chicken. The expression of myogenic factor and AChRα subunit are the same as that of nerve Implantation effects, early development AChR expression is independent of myogenin and may be regulated by other members of the MyoD family or alternative factors.
