目的:优选大孔树脂分离纯化酸枣仁醇提物中总黄酮的工艺。方法:通过静态和动态吸附-洗脱试验比较6种不同型号大孔树脂对酸枣仁总黄酮的吸附和解吸性能,筛选最佳大孔树脂型号;以总黄酮质量浓度为指标,采用单因素试验考察药液质量浓度、吸附速率、上样体积、洗脱剂浓度、洗脱速率等因素对纯化工艺的影响。结果:大孔树脂分离纯化酸枣仁总黄酮的最佳工艺条件为选择SP-207型大孔树脂,上样液质量浓度0.5 g·mL-1,吸附速率1 BV·h-1,上样量5 BV,加70%乙醇4.5 BV以4 BV·h-1的速率洗脱;酸枣仁总黄酮纯度达65.47%。结论:该优选工艺稳定可行,适用于酸枣仁总黄酮的分离纯化。
Objective: To optimize the process of separation and purification of total flavonoids from the jujube kernels by macroporous resin. Methods: The adsorption and desorption properties of total flavonoids of Semen Ziziphi spinosa L. were studied by static and dynamic adsorption-elution experiments. The best macroporous resin model was screened. The concentration of total flavonoids was used as the index, and single factor test The effects of factors such as mass concentration, adsorption rate, sample volume, eluent concentration and elution rate on the purification process were investigated. Results: The optimum conditions for the separation and purification of the total flavonoids from Spinacao jujube were as follows: SP-207 macroporous resin, the mass concentration of the sample was 0.5 g · mL-1, the adsorption rate was 1 BV · h-1, 5 BV, and 4.5 BV of 70% ethanol was eluted at the rate of 4 BV · h-1. The purity of total flavonoids of wild jujube kernel reached 65.47%. Conclusion: The optimal process is stable and feasible. It is suitable for the separation and purification of total flavonoids of wild jujube kernels.
