目的:研究异常黏液质型阳痿病证大鼠性腺轴形态学的改变。方法选用90只性功能正常的雄性SD大鼠,从中随机抽取10只设为正常对照组,其余80只为造模组,采用芫荽实+菠菜实+湿寒性环境的干预条件建立异常黏液质证候模型。动态观测大鼠生物学表征与勃起功能改变,造模20周后通过APO勃起实验和性行为学实验筛选出阳痿病证模型后,随机分为病证模型组、病证自然反证组和病证药物反证组,反证时间2周,实验终结时,取下丘脑、垂体和睾丸,HE染色,并于光镜和电镜下进行形态学观察。结果(1)下丘脑:正常组组织结构与细胞形态完好。病证模型组基质疏松,提示水肿;超微结构显示神经元核膜界线不清,线粒体水肿,内质网轻微扩张。自然反证组,部分细胞水肿变性;超微结构显示线粒体水肿,粗面内质网有轻度扩张。药物反证组,得到一定程度修复。(2)垂体:正常组结构与细胞形态完好。病证模型组基质疏松,轻度水肿,超微结构可见内质网明显扩张,线粒体普遍空泡样变。自然反证组基质亦疏松,细胞核细胞质边界不清,细胞水肿;超微结构显示大部分线粒体呈空泡样变,粗面内质网扩张。药物反证组基质疏松有所改善,水肿有所减轻。(3)睾丸:正常组结构与细胞形态完好,病证模型组可见生精小管明显变形,生精细胞层数与数量明显减少,间质减少;超微结构显示,生精细胞数量减少且排列紊乱,可见有精原细胞、精母细胞核固缩。自然反证组,生精细胞数量较少,超微结构下可见支持细胞水肿,内质网扩张呈囊泡状。药物反证组生精细胞数量上升,支持细胞增加,管腔内可见一定数量的精子。结论维医异常黏液质阳痿病证大鼠性腺轴可发生明显的病理改变,提示性腺轴的病理性改变可能是该病证发生发展的重要机制之一。“,”Objective To study the morphology changes of gonad axis in abmormal balgam syndrome rat model with impotence disease. Methods 90 mature male SD rats were selected in the study, ten of them were randomly chosen as the nomal group;the others were treated with spinach and coriander diet in a cold and humid environment. Rat biological characterization was observed about 20 weeks until the balgam syndrome model was established, then through APO erectile experiment and mating experiment,the rats were randomly divided into impotence model group,spontaneous recovering group and medication group.After 2 weeks counterevidence, the morphology of hypothalamus, pituitary and testis were observed. Results (1) The hypothalamus: The tissue structure and cell morphology of normal group was in good condition. The cell matrix of impotence model group was loose which suggested that there were swelling;ultrastructure showed the boundary of nuclear membrane was not clear, the mitochondria was edema and the rough endoplasmic reticulum was expansion. Spontaneous recovering group, parts of the cells were edema and degeneration, ultrastructure showed showed that there was slight edema in mitochondria, and mild expansion in rough endoplasmic reticulum. Medication group was a little repaired. (2) The pituitary gland: the normal group was good. The impotence model group and spontaneous recovering group, cell matrix was loose, nucleus boundary was not clear, and the cells were swelling; ultrastructure showed that most of the mitochondria was in cavitation sample change, rough endoplasmic reticulum expanded obviously. Medication group ,the matrix loose was improved and the edema was mitigated. (3) The testis: the normal group was good. The impotence model group , seminiferous ducts were obviously deformed, the number of sperm cells was decreased and disordered, some spermatagonial cells and spermatocytes were necrosis. Spontaneous recovering group, the sperm cells was less;ultrastructure showed that the support cells were swelling and endoplasmic reticulum was expanded as vesicle. Medication group,the sperm cells and supporting cells was increased, and there were some sperms in the tube cavity. Conclusion There are significant changes of in gonad axis of abnormal balgam syndrome rat model with impotence disease, which shows that pathological changes of gonad axis may be one of the mechanisms which made this syndrome occurred and developped.
