小叶黑柴胡(Bupleurum smithii Wolff var.parvifolium)总黄酮有显著的利胆退黄功效,而其如何在机体内发挥作用,目前尚未见报道.本文通过代谢物组学方法对小叶黑柴胡总黄酮防治α-萘异硫氰酸酯(α-naphthyl isothiocyanate,ANIT)所致大鼠急性黄疸型肝损伤过程中的尿液代谢组进行研究,从而探讨小叶黑柴胡总黄酮对急性黄疸型肝损伤的防治效果及作用机制.结果显示,给药组和模型组相比,大鼠尿液代谢物中马尿酸、庚酰肉毒碱、5β-胆甾烷-3α,7α,12α,23R,25-醇、4-羟双氢鞘氨醇、二氢神经鞘氨醇、溶血磷脂酰乙醇胺(0∶0/24∶0)、苯乙尿酸等含量降低,6-酮前列腺素F1α和3-磺基去氧胆酸含量升高,提示小叶黑柴胡总黄酮明显缓解α-萘异硫氰酸酯引起的大鼠急性黄疸型肝损伤,可用于防治急性黄疸型肝损伤.本文初步揭示了小叶黑柴胡总黄酮发挥药效的途径,为研究中药的作用机制提供了一个新思路.
The total flavonoids of Bupleurum smithii Wolff var.
