目的 联合检测大肠癌患者外周血细胞角蛋白 - 2 0信使核糖核酸 (CK - 2 0mRNA)与血浆中白细胞介素 - 1 0 (IL - 1 0 ) ,并评价其临床意义。方法 应用RT -PCR检测大肠癌患者外周血CK - 2 0mRNA ;应用ELISA检测其血浆IL - 1 0浓度。结果 对照组2 5例健康查体者 ,其外周血中未见CK - 2 0mRNA的表达 ;观察组 46例大肠癌患者 ,2 5例外周血中CK - 2 0mRNA表达阳性。大肠癌患者外周血CK - 2 0mRNA的表达与Dukes分期关系为 :A期 2 0 .0 % (1 / 5)、B期 2 7.3 % (3/ 1 1 )、C期 61 .1 % (1 1 / 1 8)、D期 83 .3(1 0 /1 2 ) ;其中C、D期与A、B期比较差异有高度显著性 (P <0 .0 1 )。大肠癌患者血浆IL - 1 0浓度明显高于健康查体者 ,且与Dukes分期显著相关。外周血CK - 2 0mRNA阳性表达的大肠癌患者血浆IL - 1 0水平高于阴性者。结论 联合检测大肠癌患者外周血CK -2 0mRNA与IL - 1 0 ,掌握检测大肠癌转移的相关指标 ,可以及时全面地了解大肠癌患者病情进展情况。
Objective To detect CK - 200 mRNA and IL - 10 in peripheral blood of patients with colorectal cancer and to evaluate its clinical significance. Methods RT - PCR was used to detect the CK - 2 0 mRNA in peripheral blood of patients with colorectal cancer. The plasma concentration of IL - 10 was detected by ELISA. Results There was no expression of CK - 20 mRNA in peripheral blood of 25 healthy controls in the control group. The expression of CK - 20 mRNA in 25 patients with colorectal cancer in the observation group and 25 peripheral blood was positive. The expression of CK - 20 mRNA in peripheral blood of patients with colorectal cancer was correlated with Dukes stage by 20.0% (1/5) in stage A, 7.3% (3/1) in stage B, 61.1% (1) in stage C 1/1 8), D 83.3 (1 0/1 2). The difference between C, D and A and B phases was highly significant (P <0.01). The concentration of plasma IL - 10 in patients with colorectal cancer was significantly higher than that in healthy people, and was significantly correlated with Dukes stage. The level of plasma IL - 10 in colorectal cancer patients with positive CK - 20 mRNA expression was higher than that in negative ones. Conclusions Combined detection of CK-20 mRNA and IL-1 0 in peripheral blood of patients with colorectal cancer can detect the related indicators of metastasis of colorectal cancer and can make a comprehensive and timely understanding of the progression of the disease in patients with colorectal cancer.
