目的观察姜黄素药物血清对瘦素刺激肝星状细胞增殖的影响,探讨其抗肝纤维化的机制。方法用姜黄素煎剂给大鼠灌胃(连续7 d),制备含药血清,并用培养基将药物血清配制成四个稀释倍数,分别是5%、10%、20%、40%四个稀释倍数浓度;培养肝星状细胞,取对数生长期细胞,运用噻唑蓝(MTT)比色法检测药物血清对HSC-T6增殖的影响。结果姜黄素药物血清对增殖的肝星状细胞有抑制作用(P<0.05或P<0.01),在5%~40%的血清稀释倍数范围内,随剂量增大而抑制作用增强;在20%~40%的血清稀释倍数范围内姜黄素组对HSC-T6抑制率增高(P<0.05)。结论姜黄素具有抑制HSC-T6增殖的作用。
Objective To observe the effect of serum of curcumin on the proliferation of hepatic stellate cells stimulated by leptin and to explore its mechanism of anti-hepatic fibrosis. Methods The rats were gavaged with curcumin decoction for 7 consecutive days to prepare the medicated serum. The serum was prepared with four dilutions of 5%, 10%, 20% and 40% respectively Dilution fold concentration; cultured hepatic stellate cells, logarithmic growth phase cells, the use of MTT colorimetric assay of serum on HSC-T6 proliferation. Results The serum of curcumin could inhibit the proliferation of hepatic stellate cells (P <0.05 or P <0.01), and the inhibitory effect was enhanced with the increase of dose in the range of 5% ~ 40% The inhibitory rate of HSC-T6 in curcumin group increased by ~ 40% serum dilution (P <0.05). Conclusion Curcumin inhibits the proliferation of HSC-T6.
