目的探讨凋亡相关基因survivin和p53在脑膜瘤中的表达及其临床意义。方法应用免疫组化SP法,检测97例脑膜瘤组织、18例正常脑膜组织中凋亡相关基因survivin、p53蛋白的表达,结果应用统计学分析。结果Survivin在正常脑膜组织和脑膜瘤组织中阳性表达率分别为0和65·9%(P<0·01)。Survivin表达阳性率在脑膜瘤Ⅱ级(85·7%)和Ⅰ级(51·4%)之间、Ⅲ级(93·8%)和Ⅰ级之间差异具有统计学意义(P<0·01)。p53在正常脑膜组织和脑膜瘤组织表达的阳性率分别为0和27·8%(P<0·05)。p53表达阳性率在脑膜瘤各级别之间的差异具有统计学意义(P<0·01)。Survivin与p53蛋白表达共阳性23例,共阴性29例,呈正相关(rs=0·451,P<0·01)。结论①Survivin和p53在脑膜瘤中的表达与其恶性程度相关;②Survivin基因过度表达和抑癌基因p53失活在脑膜瘤的发生和恶化中起协同作用;③Survivin将是脑膜瘤免疫治疗理想和有效的靶基因。
Objective To investigate the expression of survivin and p53 in meningiomas and their clinical significance. Methods Immunohistochemical SP method was used to detect the expression of survivin and p53 protein in 97 cases of meningioma and 18 cases of normal human meningeal tissue. The results were statistically analyzed. Results The positive rates of Survivin in normal meningioma and meningioma were 0 and 65.9% (P <0.01), respectively. The positive rate of Survivin expression in meningioma grade Ⅱ (85.7%) and grade Ⅰ (51.4%), grade Ⅲ (93.8%) and grade Ⅰ difference was statistically significant (P0.01 01). The positive rates of p53 expression in normal meningioma and meningiomas were 0 and 27.8% (P <0.05), respectively. The positive rate of p53 expression in meningioma at all levels was statistically significant (P <0.01). There was a positive correlation between Survivin and p53 protein expression in 23 cases and common negative in 29 cases (rs = 0.451, P <0.01). Conclusions ① The expression of Survivin and p53 in meningiomas correlates with their malignancy; ② Survivin gene overexpression and p53 gene inactivation play a synergistic role in the pathogenesis and progression of meningioma; ③ Survivin will be an ideal and effective target for meningioma immunotherapy. gene.
