目的:利用高效液相色谱法,建立茜草饮片的指纹图谱。方法:色谱条件,Hanbon Kromasil C18柱(4.6 mm×250mm,5μm);流动相A为乙腈,B为0.1‰三氟乙酸;洗脱方法 (0~30 min,10%~25%A;30~35 min,25%~50%A;35~55min,50%~65%A;55~70 min,65%~95%A;70~75 min,95%~10%A)。体积流量1.0 mL·min-1;柱温30℃;检测波长240nm;进样量10μL。结果:对10批茜草饮片进行测定,标定出15个共有峰,采用《中药色谱指纹图谱相识度评价系统2004A版》软件计算出其相似度范围在0.824~0.989,并得出了茜草饮片的对照指纹图谱。结论:该法准确,重复性好,可作为茜草饮片质量控制方法。
Objective: To establish the fingerprint of Rubia cordifolia by high performance liquid chromatography. Methods: The chromatographic conditions were as follows: Hanbond Kromasil C18 column (4.6 mm × 250 mm, 5 μm); mobile phase A was acetonitrile and B was 0.1 ‰ trifluoroacetic acid; the elution method was 0 ~ 30 min, 10% ~ 25% 35 min, 25% -50% A, 35-55 min, 50% -65% A, 55-70 min, 65% -95% A, 70-75 min, 95% -10% A). Volume flow 1.0 mL · min-1; column temperature 30 ℃; detection wavelength 240nm; injection volume 10μL. Results: Ten batches of Rubia cordifolia samples were determined and 15 common peaks were calibrated. The similarity between the two samples was 0.824 ~ 0.989 using the software of Chinese Medical Chromatography Fingerprint Evaluation System 2004A, and the control Fingerprint. Conclusion: This method is accurate and reproducible, and can be used as a quality control method for Rubia cordifolia.
