目的观察普伐他汀对醛固酮诱导新生大鼠心脏成纤维细胞转化生长因子βⅠ型受体(TGF-βRI) mRNA表达的影响及其机制。方法采用胰酶消化法和差速贴壁分离法获取新生大鼠心脏成纤维细胞,应用RT-PCR的方法测定TGF-βR I mRNA的表达。结果醛固酮(0.1μmol·L~(-1))可诱导心脏成纤维细胞中TGF-βR I mRNA的表达,预先24 h给予普伐他汀(0.01、0.1、1 mmol·L~(-1))能剂量依赖性抑制醛固酮的上述作用,同时,普伐他汀的这种抑制作用可被甲羟戊酸所逆转。结论普伐他汀可抑制醛固酮诱导的心脏成纤维细胞TGF-βR I mRNA表达,其机制可能与甲羟戊酸代谢途径有关。
Objective To investigate the effect and mechanism of pravastatin on aldosterone-induced TGF-βRI mRNA expression in neonatal rat cardiac fibroblasts. Methods Newborn rat cardiac fibroblasts were obtained by trypsin digestion and differential adherent separation. The expression of TGF-βR I mRNA was determined by RT-PCR. Results Aldrin (0.1μmol·L -1) induced the expression of TGF-βR I mRNA in cardiac fibroblasts. Pravastatin (0.01,0.1,1 mmol·L -1) This dose-dependent inhibition of aldosterone, while pravastatin this inhibition can be reversed by mevalonate. Conclusions Pravastatin can inhibit aldosterone-induced TGF-βR I mRNA expression in cardiac fibroblasts, which may be related to the metabolic pathway of mevalonate.
