以秀丽隐杆线虫模式生物为研究对象,研究大豆异黄酮染料木素微生物代谢产物左旋-5-羟基-雌马酚对线虫寿命、生殖能力以及抗热应激和抗氧化应激能力的影响。研究结果表明,浓度为0.1 mmol/L和0.2 mmol/L的左旋-5-羟基-雌马酚不仅能够显著延长线虫寿命,同时还能显著增强线虫耐热和抗氧化应激能力,而在相同浓度下线虫生殖能力则无明显不良影响。通过进一步对比分析转录物水平以及喂食相应线虫突变体daf-16(mu86)和daf-2(e1370),确定左旋-5-羟基-雌马酚延长线虫寿命与DAF-2/DAF-16 Insulin/IGF-1信号转导通路相关。有关左旋-5-羟基-雌马酚显著延长线虫寿命和增强线虫抗性为国内外首次报道。
The effects of L-5-hydroxy-equol, a metabolic product of soy isoflavone genistein, on the lifespan, reproductive capacity, anti-heat stress and anti-oxidative stress of C. elegans were studied. The results showed that the concentration of 0.1 mmol / L and 0.2 mmol / L of L-5-hydroxy-equol not only can significantly prolong the life of nematodes, but also significantly enhances the ability of heat-tolerance and antioxidant stress of nematodes, The concentration of nematode reproductive ability without obvious adverse effects. DAF-2 / DAF-16 Insulin / DAF-16 was further identified by comparing the transcript levels with the corresponding nematode mutants daf-16 (mu86) and daf-2 IGF-1 signaling pathway. About L-5-hydroxy-equol significantly prolonged nematode life and enhanced nematode resistance for the first time reported at home and abroad.
