目的研究纤溶酶原激活物(uPA)与基质金属蛋白酶(MMP-9)在大鼠矽肺纤维化发生发展中的意义。方法建立大鼠矽肺模型组及对照组,分别取不同时间点处死大鼠,取其肺组织,免疫组化法检测肺组织中uPA、MMP-9蛋白的表达并分析其相关性。结果与对照组比较,uPA的阳性表达随时间延长呈增强趋势,第1周时达高峰,此后逐渐减弱,但仍较对照组明显。MMP-9阳性表达强度随染尘时间延长而逐渐增强,至第7天时达高峰,此后呈减弱趋势。uPA、MMP-9蛋白表达与对照组比较,差异有统计学意义(P<0.05)。其显色指数与对照组比较,差异有统计学意义(P<0.05)。uPA、MMP-9蛋白表达呈正相关。结论肺组织内uPA、MMP-9蛋白表达在肺纤维化过程中发挥重要的作用,可能因为uPA诱导的MMPs活化与其纤溶活性共同改变了细胞外基质胶原的沉积,从而影响矽肺纤维化的发生发展。
Objective To study the significance of uPA and MMP-9 in the occurrence and development of silicotic fibrosis in rats. Methods Silicosis model group and control group were established. The rats were sacrificed at different time points, the lung tissues were taken out and the expressions of uPA and MMP-9 protein in lung tissue were detected by immunohistochemistry. Results Compared with the control group, the positive expression of uPA tended to increase with time and peaked at the first week, then gradually weakened, but still more obvious than the control group. The positive expression intensity of MMP-9 gradually increased with the prolongation of dust exposure, reached its peak on the 7th day, and then decreased. The expression of uPA and MMP-9 protein were significantly different from the control group (P <0.05). The color rendering index compared with the control group, the difference was statistically significant (P <0.05). uPA, MMP-9 protein expression was positively correlated. Conclusion The expression of uPA and MMP-9 in lung tissue play an important role in the process of pulmonary fibrosis. The possible mechanism is that uPA-induced activation of MMPs and its fibrinolytic activity may alter the deposition of extracellular matrix collagen and thus affect the occurrence of silicotic fibrosis development of.
