为研究胭脂萝卜花青素的提取工艺,考察了液料比、浸提温度、浸提时间、超声功率、超声时间对花青素提取率的影响。采用MTT实验、细胞侵袭实验研究了花青素对人胃癌细胞NCIN87增殖、侵袭的作用,进一步用免疫印迹分析其对HER2信号通过的影响。结果表明,结合响应面分析得出胭脂萝卜花青素最佳提取工艺为:以1%盐酸乙醇为提取剂,液料比10∶1,40℃浸提2h,400W超声波破碎15min,在此条件下,花青素的最高提取量为3.92mg/g。30μg/ml的胭脂萝卜花青素能显著抑制NCI-N87细胞的增殖和侵袭能力,同时抑制HER2蛋白及Akt的磷酸化水平。
In order to study the extraction technology of anthocyanin in carmine turnip, the effects of liquid ratio, extraction temperature, extraction time, ultrasonic power and ultrasonic time on the extraction rate of anthocyanins were investigated. The effects of anthocyanin on the proliferation and invasion of human gastric cancer cell NCIN87 were studied by MTT assay and cell invasion assay. The effect of anthocyanin on HER2 signaling was further analyzed by Western blotting. The results showed that the optimal extraction process of anthocyanin from Radix Bupleuri was as follows: extracting with 1% hydrochloric acid and ethanol as solvent, leaching at 10: 1, 40 ℃ for 2 h, then sonicating with 400 W ultrasound for 15 min, The maximum amount of anthocyanin extraction was 3.92 mg / g. The carrageenan at 30μg / ml could significantly inhibit the proliferation and invasion of NCI-N87 cells and inhibit the phosphorylation of HER2 protein and Akt.
