目的:探讨肿瘤坏死因子相关凋亡诱导配体(TRAIL)信号途径在多囊卵巢综合征(PCOS)大鼠卵巢颗粒细胞诱导凋亡的最终效应阶段的作用。方法:采用硫酸普拉睾酮钠诱导大鼠PCOS模型,采用Western blot及RT-PCR分析半胱氨酸天门冬氨酸蛋白酶3(Caspase3)在PCOS大鼠卵巢颗粒细胞的表达情况。结果:①Western blot分析显示,PCOS组大鼠卵巢颗粒细胞活性Caspase3蛋白的表达较对照组明显增强(P<0.01)。②RT-PCR分析显示,PCOS组大鼠卵巢颗粒细胞Caspase3mRNA的表达较对照组明显增强(P<0.01)。结论:TRAIL信号途径在PCOS大鼠颗粒细胞诱导的凋亡可能最终经过了Caspase级联反应,引起Caspase3的激活导致细胞凋亡。
AIM: To investigate the role of tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) signaling pathway in the final induction of apoptosis in ovarian granulosa cells of rats with polycystic ovary syndrome (PCOS). Methods: Rat model of PCOS was induced by sodium pramipexate sulfate. The expression of Caspase3 in ovarian granulosa cells of PCOS rats was analyzed by Western blot and RT-PCR. Results: (1) Western blot analysis showed that the expression of Caspase3 protein in ovarian granulosa cells in PCOS group was significantly higher than that in control group (P <0.01). ② RT-PCR analysis showed that the expression of Caspase3mRNA in ovarian granulosa cells in PCOS group was significantly higher than that in control group (P <0.01). CONCLUSION: Apoptosis induced by TRAIL signaling in granulosa cells of PCOS rats may eventually undergo Caspase cascade, leading to activation of Caspase3 resulting in apoptosis.
