体外观察干扰素 γ(interferon γ ,IFN γ)对脐血造血干 /祖细胞细胞周期蛋白D (cyclinD)不同亚型表达的影响 ,以探讨IFN γ造血抑制的机制。用免疫磁珠阳性分选的方法分离脐带血CD34+细胞 ,并进行体外扩增 ;用甲基纤维素半固体培养方法 ,观察IFN γ对脐血造血干 /祖细胞CFU GM形成的影响 ;用半定量RT PCR检测IFN γ对脐血造血干 /祖细胞cyclinD不同亚型表达的影响。结果显示IFN γ可以显著抑制脐血CFU GM的形成 ,并可以从mRNA水平抑制cyclinD2和CyclinD3亚型的表达。由此得出结论 ,IFN γ可以显著抑制cyclinD的表达 ,这可能是IFN γ介导造血抑制的一种机制。
To observe the effect of interferon γ (IFNγ) on the expression of cyclinD subunit of cord blood hematopoietic stem / progenitor cells in vitro, in order to investigate the mechanism of IFN γ hematopoietic inhibition. Umbilical cord blood CD34 + cells were isolated by immunomagnetic bead positive sorting and expanded in vitro. The effects of IFNγ on the CFU GM formation of umbilical cord blood hematopoietic stem / progenitor cells were observed by methylcellulose semi-solid culture method. Effect of IFNγ on the expression of cyclinD in different types of cord blood hematopoietic stem / progenitor cells by quantitative RT - PCR. The results showed IFNγ could significantly inhibit the formation of CFU GM in umbilical cord blood and inhibit the expression of cyclinD2 and CyclinD3 subtypes from mRNA level. It was concluded that IFNγ could significantly inhibit the expression of cyclinD, which may be a mechanism of IFNγ-mediated inhibition of hematopoiesis.
