Objective:To studythevacuolatingeffectof Helicobacter pylori(H.pylori).Method:Thevacuolatingef-fectanditsrelationshipwithvacuolatingcytotoxinantigen(an87kDprotein)wereinvestigatedby themethodof cy-totoxictest,SDS-PAGEandscanning.Result:Of the62clinicalisolates,43strainswereH.pylori(Toxin+)with vacuolatingeffect,whiletheotherswereH.pylori(Toxin-)withoutvacuolatingeffect.Altogether78.26%(36/46)patientswithpepticulcerwereinfectedwithH.pylori(Toxin+)strains,andonly42.86%(6/14)who hadgastritiswere infectedwithH.pylori(Toxin+)strains,withsignificantdifferencebetweenthem(χ 2 =4.83,P<0.05).A proteinwith relativemolecularmassof87kD was identifiedin thebrothculturefilter(BCF)of30.23%H.pylori(Toxin+)strains(13/43)butinnoneof thatof H.pylori(Toxin-)strains,andthedifferencewas statisticallysignificant(P<0.05).There was a significantandconcordantrelationshipbetweentheOD valueof theproteinbandandthetiterof vacuolating activityof H.pylori(Toxin+)(r=0.67andP<0.05by linearregressionanalysis).Conclusion:H.pylori(Toxin+)were moreoftenassociatedwithpepticulcerousdiseasesthanwithgastritisdiseases.Thevacuolatingeffectof H.pylori(Toxin+)maybe causedby the87kDprotein.
