目的 探讨bcl-xS基因对化疗后大肠癌细胞生物学行为的影响。方法 用基因转染方法进行LoVo细胞bcl-xS基因转染,然后用MTT和ELISA法检测bcl-xS转染细胞对化疗药MMC及5-FU敏感性的影响和作用机制。结果 bcl-xS转染可增强LoVo细胞对化疗药MMC及5-FU的敏感性,与经neo转染的细胞相比有显著差异(P<0.05)。ELISA细胞死亡试剂盒研究发现,LoVo细胞对化疗药敏感性的增加是由于凋亡所致,而且MMC和5-FU对bcl-xS转染细胞凋亡的诱导是对照组neo转染细胞的1~4倍。结论 本研究结果提示了bcl-xS转染途径的靶治疗功用。
Objective To investigate the effect of bcl-xS gene on the biological behavior of colorectal cancer cells after chemotherapy. Methods The bcl-xS gene was transfected into LoVo cells by gene transfection. Then the effect of bcl-xS transfected cells on the sensitivity of chemotherapeutic agents MMC and 5-FU and its mechanism were detected by MTT and ELISA. Results The bcl-xS transfection enhanced the sensitivity of LoVo cells to chemotherapeutic agents MMC and 5-FU, which were significantly different from those transfected with neo (P <0.05). ELISA cell death kit study found that LoVo cells increased sensitivity to chemotherapeutic drugs is due to apoptosis caused by MMC and 5-FU on bcl-xS transfected cells apoptosis induced neo-transfected cells in the control group 1 ~ 4 times. Conclusion The results of this study suggest the target therapeutic efficacy of the bcl-xS transfection route.
