锶矿泉水对人血管内皮细胞的增殖和功能的影响

目的观察不同浓度的锶矿泉水对人脐静脉血管内皮细胞(HUVEC)增殖及分泌的影响,研究其对血管内皮细胞功能的作用。方法 HUVEC细胞分为2、4、6、8 mg/L锶矿泉水(SMW)组和双蒸水(DDW)组,以4×104个/ml的浓度接种于培养板中,18 h后,换成条件培养基培养至96 h。分别于24、48、72和96 h时,以细胞计数试剂盒-8(CCK-8)法观察细胞的增殖情况。培养72 h的细胞分别用苏木素染色后,观察细胞生长状态,以及用BCA蛋白化验试剂盒测蛋白含量,酶联免疫吸附试验(ELISA)检测培养基中血管内皮生长因子(VEGF)、一氧化氮(NO)和内皮素(ET)的含量。结果培养72 h后,2和4 mg/L SMW组细胞增殖速度快于DDW组(P<0.05);8 mg/LSMW抑制HUVEC增殖(P<0.05);培养至72 h时,各浓度的SMW组细胞分泌VEGF的量较DDW组均降低(P<0.05),其组间差异无统计学意义(P>0.05);2和4 mg/L SMW组分泌ET和NO减少(P<0.05),而6和8 mg/LSMW组分泌增多(P<0.05),但同一浓度下,ET和NO分泌变化呈正相关。4 mg/L SMW组NO/ET值增大(P<0.05),其余各组间差异无统计学意义(P>0.05)。结论一定浓度的锶矿泉水可促进血管内皮细胞增殖,影响内皮细胞ET和NO分泌,降低血管紧张性。VEGF并不直接参与锶矿泉水促进血管内皮细胞的增殖。 Objective To observe the effects of different concentrations of strontium mineral water on the proliferation and secretion of human umbilical vein endothelial cells (HUVECs) and to study their effects on the function of vascular endothelial cells. Methods HUVECs were divided into 2,4,6,8 mg / L strontium mineral water (SMW) group and double distilled water (DDW) group and inoculated into the culture plate at the concentration of 4 × 104 / ml. Results After 72 h culture, the cell proliferation rate in 2 and 4 mg / L SMW group was faster than that in DDW group (P <0.05), and 8 mg / LSMW inhibited the proliferation of HUVEC (P <0.05) Compared with DDW group, the amount of VEGF secreted by group 2 cells was lower than that of DDW group (P <0.05), and there was no significant difference between groups (P> 0.05) However, secretion of 6 and 8 mg / LSMW groups increased (P <0.05), but there was a positive correlation between ET and NO secretion at the same concentration. NO / ET in 4 mg / L SMW group increased (P <0.05), but there was no significant difference among other groups (P> 0.05). Conclusion Certain concentration of strontium mineral water can promote the proliferation of vascular endothelial cells, affect the secretion of ET and NO in endothelial cells and reduce the vascular tone. VEGF does not directly participate in strontium mineral water to promote the proliferation of vascular endothelial cells.
