目的考察8种不同规格鹿茸含药血清对MC3T3-E1成骨细胞增殖和分化的影响并对8种不同规格鹿茸进行聚类分析。方法雌性SD大鼠随机分为9组,给药,制得不同规格鹿茸的大鼠含药血清及空白对照血清。以上述血清对MC3T3-E1成骨细胞培养72 h,MTT法检测成骨细胞增殖情况,ELISA法检测碱性磷酸酶(ALP)分泌量,并以成骨细胞的增殖及ALP分泌量为因素进行聚类分析。结果各规格鹿茸均具有促进MC3T3-E1成骨细胞增殖、分化作用,且聚类分析结果与传统商品鹿茸药材质量划分结果相同。结论鹿茸药材能显著促进MC3T3-E1成骨细胞增殖、分化,且作用强弱与商品药材优劣相关。
Objective To investigate the effects of eight different antler drug-containing serums on proliferation and differentiation of MC3T3-E1 osteoblasts and cluster analysis of eight different types of antler. Methods Female Sprague-Dawley rats were randomly divided into 9 groups, administered with drug-containing serum of different specifications of pilose antlers and blank control serum. The above serum was used to culture MC3T3-E1 osteoblasts for 72 h. The proliferation of osteoblasts was detected by MTT assay. The alkaline phosphatase (ALP) secretion was detected by ELISA. The factors of osteoblasts proliferation and ALP secretion Cluster analysis. Results The velvet antlers all promoted the proliferation and differentiation of MC3T3-E1 osteoblasts, and the results of cluster analysis were the same as those of traditional commercial pilose antlers. Conclusion Pilose antler can significantly promote the proliferation and differentiation of MC3T3-E1 osteoblasts, and its effect is related to the quality of commercial medicinal herbs.
