目的:探讨促红细胞生成素产生肝细胞B6(EPHB6)对于胞外信号调节激酶(ERK)信号转导通路的作用.方法:以非小细胞肺癌细胞系A549作为研究对象,采用Western Blot法检测ERK的磷酸化水平及通路检测(PathDe-tect)法检测ERK下游分子和Ets结构域蛋白1(Elk-1)转录因子的活性.结果:A549细胞中EPHB6的表达导致ERK的磷酸化.相反,siRNA干涉EPHB6的表达可以逆转ERK的磷酸化.并且,EPHB6导致的ERK磷酸化与Elk-1转录因子脱偶联.结论:表明EPHB6对于ERK信号通路具有调控作用.
Objective: To investigate the effect of EPHB6 on the extracellular signal-regulated kinase (ERK) signal transduction pathway.Methods: The non-small cell lung cancer cell line A549 was used as the research object, Western Blot method was used to detect the expression of ERK Phosphorylation and PathDe-tectometry.Results: The expression of EPHB6 in A549 cells resulted in the phosphorylation of ERK.Conversely, siRNA Interference of EPHB6 expression reversed the phosphorylation of ERK, and the phosphorylation of ERK induced by EPHB6 was uncoupled with Elk-1 transcription factor.Conclusion: EPHB6 can regulate ERK signaling pathway.
