目的探讨人参皂甙RH2(G-RH2)对人肺腺癌A549细胞凋亡的影响。方法分析G-RH2对A549细胞周期及细胞凋亡的影响,并检测G-RH2对A549细胞相关基因表达的影响。结果 0~60μmol的G-RH2作用A549细胞72 h后,实验组G1期细胞比率随药物浓度的增加而增加,S期细胞比率随药物浓度的增加而递减,G2期细胞无明显变化。Bax、Bid及Cyto-C表达量随药物浓度增加而增加,Bcl-2、细胞外信号调节激酶(ERK)及P-ERK表达量随药物浓度的增加而降低。结论 G-RH2可以诱导人肺腺癌A549细胞凋亡,G-RH2抗肿瘤药物值得临床开发和应用。
Objective To investigate the effect of ginsenoside RH2 (G-RH2) on the apoptosis of human lung adenocarcinoma A549 cells. Methods The effect of G-RH2 on A549 cell cycle and apoptosis was analyzed and the effect of G-RH2 on A549 cell-related gene expression was examined. Results After treated with 0 ~ 60μmol G-RH2 for 72h, the percentage of cells in G1 phase of experimental group increased with the increase of drug concentration. The percentage of cells in S phase decreased with the increase of drug concentration, but there was no significant change in G2 phase. The expression of Bax, Bid and Cyto-C increased with the increase of the drug concentration. The expression of Bcl-2, extracellular signal-regulated kinase (ERK) and P-ERK decreased with the increase of the drug concentration. Conclusion G-RH2 can induce apoptosis of human lung adenocarcinoma A549 cells, and G-RH2 antitumor drugs deserve clinical development and application.
