本文应用体外软琼脂培养技术,以PHA-LCM条件培养液作为白血病细胞集落形成刺激因素,对10例急性粒细胞白血病患者进行了L-CFU培养。结果10例患者的骨髓细胞均有L-CFU形成。培养7天后L-CFU形成达到高峰。每1×10~6骨髓细胞可形成74~451个集落,细胞丛197~1,015个。7例患者进行了外周血L-CFU培养,结果亦都有L-CFU形成。L-CFU数为26~700个,细胞丛83~976个。为了解L-CFU组成细胞性质,进行了培养前后的细胞化学染色及电镜等观察,结果表明,培养后所形成集落的组成细胞均出现幼稚细胞特征。2例在培养前后的幼稚细胞中均见到了Auer小体。从上述观察推定集落来源于白血病细胞。
In this paper, soft agar culture in vitro techniques, PHA-LCM conditioned medium as a leukemic cell colony stimulating factor, 10 cases of acute myeloid leukemia in L-CFU culture. Results All 10 patients had L-CFU formation. L-CFU reached its peak after 7 days of culture. There are 74 ~ 451 colonies per 1 × 10 ~ 6 bone marrow cells with 197 ~ 1,015 cells. Seven patients had peripheral blood L-CFU culture, the results also have L-CFU formation. The number of L-CFU was 26 to 700, and the number of cell clusters was 83 to 976. To understand the nature of L-CFU cells, cytochemical staining and electron microscopy were performed before and after culturing. The results showed that immature cells were found in all the constituent cells of the colonies after culture. Auer bodies were seen in 2 cases of naive cells before and after culture. It is presumed from the above observation that the colony is derived from leukemia cells.
