现代药理动力学包括线性和非线性两大部分。前者发展较早,乃是经典部分,而后者则是现代药理动力学的重要进展。本文对非线性药理动力学几个基本问题的研究,作了系统的介绍,我相信对于读者会有裨益。线性与非线性药理动力学有着本质不同的特点,故原则上不许混淆。就药物与机体之间相互作用的本来面貌而言,纯粹的线性动力学是罕见的,大多含有非线性成分;然而,在有效药物浓度范围内,若非线性动力学实际上可作线性处理,便会给临床用药带来极大的方便。事实上,不仅药理动力学有线性和非线性的区别。近代理论生物学指出,生命结构是个非平衡的动力体系,它在常温下不断地与外界交换着物质和能量,自我复制和自我组织。此种结构是在远离平衡态依靠耗散过程而形成和维持的,其中非线性项,例如自催化和自复制是本质的。
