目的:观察不同粒径的参附汤超微粉单次给药在大鼠体内的毒代动力学研究,评价其安全性,为临床研究提供依据。方法:采用高效液相色谱法测定参附汤传统饮片与超微粉单次给药后血浆中乌头碱的含量来研究乌头碱在大鼠体内的毒代动力学过程。结果:参附汤超微粉中乌头碱在大鼠体内呈一级动力学消除,具有开放二房室模型的特征。结论:参附汤超微粉符合急救用药的特性,可作为临床急救用药;但有体内蓄积的可能,不可久服。
OBJECTIVE: To observe the toxicokinetic dynamics of a single dose of Shenfu Decoction with different particle sizes in rats, evaluate its safety, and provide evidence for clinical research. Methods: The content of aconitine in plasma of Shenfutang traditional decoction pieces and ultramicro powder after single administration was determined by high performance liquid chromatography to study the toxicokinetic process of aconitine in rats. RESULTS: The aconitine in Shenfu Decoction was eliminated by first-order kinetics in rats, which was characterized by an open two-compartment model. Conclusion: Shenfu decoction is suitable for the first-aid medication, but it can be used as a clinical first-aid medication;
